I have turned off opportunistic locking, temporarily disabled anti virus, made sure that only one computer is opening the data at a time, and the backup is scheduled to run at night.

I tested transfer speeds from the NAS and they are getting full gigabit speed. Only when opening files from the diskstation is there a problem.

ArcGIS will open a project file stored on another windows file share just fine. They have also reported that working with files from the NAS is much slower than working from files stored on another workstation.

Possible problems can include bad network connection, invalid field, etc. As objects in a geodatabase are locked, access to these objects changes. If youre Geodatabase is still locked after running that tool, someone is editing it (hopefully you if its just a File GDB).
